Zonisamide antiepileptic drug and its pharmacology

INTRODUCTION:-
* The compound belongs to the class of organic compounds known as benzisoxazoles.
*  These are aromatic compounds containing a benzene ring fused to an isoxazole ring.

MECHANISM OF ACTION :-
* It binds to sodium channels and voltage sensitive calcium channels,which suppresses neuronal depolarization .
* It also inhibits carbonic anhydrase to a weaker extent, but such an effect is not thought to contribute substantially to the drug’s anticonvulsant activity.

PHARMACOKINETICS :-
* VOLUME OF DISTRIBUTION :- 1.45L/Kg
* PROTEIN BINDING :-40%
* HALF LIFE :-63 hrs.
* ELIMINATION :- Through urine.

SIDE EFFECTS :-
* Loss of consciousness.
* Low blood pressure.
* Slow heart beat.

DRUG-DRUG INTERACTIONS:-
* DICLOFENAMIDE+ ZONISAMIDE=Severity of adverse effect can be increased.
* DASATINIB+ZONISAMIDE=Serum concentration of zonisamide can be increased.

THERAPEUTIC USES:-
* For use as adjuvant treatment of partial seizures in adults with epilepsy
