Loratidine-antihistaminic drug

Loratidine

Mechanism of action
it acts as a selective inverse agonist of peripheral histamine H1 receptors . it blocks the action of endogenous histamine, which subsequently leads to temporary relief of the negative symptoms brought on by histamine.

Drug drug interaction
Loratidine+ketoconazole = increased plasma level of loratidine
Loratidine+erythromycin = increased plasma level of loratidine
Loratidine+Cimetidine = increased plasma level of loratidine
Loratidine+aminodarone = serum concentration of loratidine is increased
Loratidine+amphetamine = decreases sedative activity of loratidine

Therapeutic uses
Sympatomatic relief of allergy
In urticaria
Effective in nasal and eye symptoms

Side effects
Headache
Urinary retention
Dry mouth
Blurred vision
Gastrointestinal disturbances

Pharmacological actions
It is a second generation peripheral histamine H1 receptor blocker used to treat allergies. In structure it is closely related to TCA and is distantly related to atypical antipsychotic quetiapine.
