Flucytosine-Antifungal drug and its pharmacology

MECHANISM OF ACTION
It acts directly on fungal organisms by competitive inhibition of purine and pyrimidine uptake and indirectly by intracellular metabolism to 5fluorouracil.

 THERAPEUTIC USES

  • Used for treatment of serious infection caused from strains of candida or Cryptococcus neoformans
  • Also used for treatment of chromomycosis.

SIDE EFFECTS 

  • Anemia 
  • Leukopenia
  • Agranulocytosis 
  • Dudonal ulcer
  • Bone marrow toxicity

CONTRAINDICATION

  • Concomitant treatment with brivudine is an absolute contraindication.

DRUG DRUG INTERACTION
1) Flucytosine+clozapine=risk of flucytosine adverse effect isincreased
2) Fucytosin+amphotericin B= risk of flucytosin adverse effect is increased
