MECHANISM OF ACTION
It interacts with 14 alpha demethylase,a CYP450 enzyme necessary to convert lanosterol to ergosterol
As ergosterol is an essential component of fungal cell membrane ,inhibittion of its synthesis results in increased cellular permeability causing lekage of cellular contents.
THERAPEUTIC USES
- Used as an anti fungal medication
- Also sometimes used in treatment of non systemic candida infection of vagina, throat and mouth.
SIDE EFFECTS
- Nausea
- Vomiting
- Anorexia
- Alopecia
- hypocalemia
CONTRAINDICATION
- Contra indicated in patients who have hypersensitivity to other azole medicines such as ketoconazole.
DRUG DRUG INTERACTION
1)Fluconazole+aminophylin=metabolism of amino phylin is decreased
2)Fluconazole+ amitriptylline= amitriptylline increase the QTc-prolonging activities of fluconazole
3)Fluconazole+amiodarone = metabolism of amiodarone decrease.
