Nitrazepam-hypnotic drug and its pharmacology

NITRAZEPAM-
A benzodiazepine derivative used as an anticonvulsant and hypnotic.

Pharmacology:
Indication- Used to treat short-term sleeping problems (insomnia), such as difficulty falling asleep, frequent awakenings during the night, and early-morning awakening.

Pharmacodynamics-Nitrazepam is a type of benzodiazepine drug. It is a powerful hypnotic drug which possesses strong sedative, anxiolytic, amnestic, anticonvulsant, and skeletal muscle relaxant properties.  Nitrazepam shortens the time required to fall asleep and lengthens the duration of sleep. It is also useful for the management of myoclonic seizures.

Absorption-Bioavailability is 53-94% following oral administration.

Half life-15-38 hours (mean elimination half life 26 hours).

Drug Drug interactions:
Azelastine+Nitrazepam= Nitrazepam may increase the central nervous system depressant (CNS depressant) activities of Azelastine.
Baclofen+Nitrazepam= The risk or severity of adverse effects can be increased when Baclofen is combined with Nitrazepam.
Cosyntropin+nitrazepam=Cosyntropin may increase the hepatotoxic activities of Nitrazepam.
Doxylamine+Nitrazepam=Doxylamine may increase the central nervous system depressant (CNS depressant) activities of Nitrazepam.
Ethanol+Nitrazepam=Nitrazepam may increase the central nervous system depressant (CNS depressant) activities of Ethanol.

Food Interactions
  >Avoid alcohol.
  >Avoid excessive quantities of coffee or tea (Caffeine).
  >Take with food.

Therapeutic Uses:
>It is used for short-term treatment of sleeping problems (insomnia), such as difficulty falling asleep, frequent awakenings during the night, and early-morning awakening.
>It is also used to manage myoclonic seizures.
>It works by slowing down the nerves in the brain (the central nervous system).

Side effects:
> clumsiness or unsteadiness
> dizziness
> drowsiness
> dry mouth
> false sense of well-being
> increased watering of mouth
>  lightheadedness
> vivid dreams
