• Lamivudine is a synthetic nucleoside analogue of cytidine.

• It’s phosphorylating to its active 5'-triphosphate metabolite, lamivudine triphosphate (L-TP).
• It inhibits type1 and type2 of HIV reverse transcriptase and the reverse transcriptase of hepatitis B virus as well.
• It is phosphorylated to active metabolites that compete for incorporation into viral DNA.
• They competitively inhibit HIV reverse transcriptase enzyme and terminates DNA synthesis.
• In result the lack of 3’-OH group in the nucleoside analogue prevents the formation of the 5' to 3' phosphodiester linkage essential for DNA chain elongation, and thus the viral DNA growth is stoppedted.
